M9 Minimal Media

Per liter of sterile water:

  • 5X M9 salts* 200 mL
  • 1M MgSO4 2 mL
  • 20% glucose** 20 mL
  • 1M CaCl2 0.1 mL

5X M9 salts

Per liter:

  • Na2HPO4 64 grams
  • KH2PO4 15 grams
  • NaCl 2.5 grams
  • NH4Cl 5 grams

In this recipe, 15N ammounium chloride and 13C glucose can be used to label proteins.

Glucose cannot be sterilized in the autoclave. It must be passed through a ~0.2μm membrane to be sterilized.
