Choosing a Plasmid

This step is necessary if your protein target does not come with a designated plasmid. Instead of steps, these are requirements for what should be located on your plasmid.

  1. Protein target sequence inserted
  2. N- or C-terminal histidine tag of varying length
  3. Antibiotic selection sequence- ampicillinm, kanamycin, or chloramphenicol.
  4. Optional- his tag cleavage site. Either TEV or thrombin.
  5. Optimized for expression in E. coli.
